Yeah I think only want some new gear because I can.

So going to think about it all.

I thought going from a Squier MIM Fender would a big upgrade.

Fender Squier AffinityHSS Stratocaster
Vintage V100, Epiphone Dove Pro
Fender Mustang 1, Peavey Vypyr 30

I think it depends on how you define a big upgrade. I never tried a Squier but I do have a MIM Fender (Tele) which I am very(!) happy with.
It may be a big upgrade to some and a minor upgrade to others..... Not much else to do than to try a MIM out and decide for yourself if the jump is big enough.

Hardware wise, the quality jump is less than if you just buy new pickups, tuners etc from (fill in brands of choice here), because the Squier Affinity's pickups for instance are just about the same as the MiM ones.
The wood might be a different story, but whether it is worth spending the extra cash on...
Maybe you can trade in the Squier and get a nice deal on the MiM ?
Then it's easily justified.

Would it be better saving a bit more money to buy MIJ or MIA?

If it were me, I'd go for one of those amps you listed in your initial post.

If you really want to make a jump in quality that can be justified, then yes.
